Chromosome polymorphism in the Brazilian dwarf brocket deer, Mazama nana (Mammalia, Cervidae) Genet. Mol. Biol.
Abril,Vanessa Veltrini; Duarte,José Maurício Barbanti.
The Brazilian dwarf brocket deer (Mazama nana) is the smallest deer species in Brazil and is considered threatened due to the reduction and alteration of its habitat, the Atlantic Rainforest. Moreover, previous work suggested the presence of intraspecific chromosome polymorphisms which may contribute to further population instability because of the reduced fertility arising from the deleterious effects of chromosome rearrangements during meiosis. We used G- and C-banding, and nucleolus organizer regions localization by silver-nitrate staining (Ag-NOR) to investigate the causes of this variation. Mazama nana exhibited eight different karyotypes (2n = 36 through 39 and FN = 58) resulting from centric fusions and from inter and intraindividual variation in...

Comparative cytogenetic analysis in the species Uroderma magnirostrum and U. bilobatum (cytotype 2n = 42) (Phyllostomidae, Stenodermatinae) in the Brazilian Amazon Genet. Mol. Biol.
Silva,Adailton Moreira da; Marques-Aguiar,Suely Aparecida; Barros,Regina Maria de Souza; Nagamachi,Cleusa Yoshiko; Pieczarka,Julio Cesar.
The genus Uroderma includes two species: U. magnirostrum and U. bilobatum. These species are characterized by their high degree of karyotypic evolution, diverging from most other species of the subfamily Stenodermatinae, which have a lower degree of chromosomic evolution. The present study reports the first banding patterns of U. magnirostrum (G-, C-banding and Ag-NOR) and U. bilobatum (C-banding and Ag-NOR). The chromosomic data in conventional staining of U. magnirostrum (2n = 36, NF = 62) and U. bilobatum (cytotype 2n = 42, NF = 50) are equivalent to that described in the literature. When compared, chromosomal homeologies are found in both karyotypes, as well as differences, confirming that karyotypic evolution in the Uroderma genus is intense. Fission,...

Comparative cytogenetic studies of Bufo ictericus, B. paracnemis (Amphibia, Anura) and an intermediate form in sympatry Genet. Mol. Biol.
Azevedo,MFC; Foresti,F.; Ramos,PRR; Jim,J.
Specimens of Bufo ictericus, Bufo paracnemis and a third type, considered an intermediate subgroup between these species, were cytogenetically studied by conventional Giemsa staining, C-banding and staining of the nucleolus organizer region (NOR). The nuclear DNA content and seroproteins were also analyzed to characterize these species, and verify the possibility of hybridization between them. Karyotypes and cytogenetic markers were essentially equal on the basis of the methods used. The DNA nuclear content found was 6.25 ± 0.30 pg/DNA in Bufo ictericus; 7.57 ± 0.40 pg/DNA in Bufo paracnemis and 7.04 ± 0.29 pg/DNA in the intermediate subgroup. Eletrophoresis of total blood serum in Bufo ictericus, Bufo paracnemis and the intermediate specimens revealed a...

Comparative karyotype analysis in diploid and triploid Dolichoplana carvalhoi (Tricladida, Terricola, Rhynchodemidae) from Brazil Genet. Mol. Biol.
Alvarez,Luciana; Almeida,Eunice Judith Cardoso de.
In this work, we present cytogenetic data for the land planarian Dolichoplana carvalhoi. Two different karyotypes, one diploid (2n = 2x = 14 chromosomes) and one triploid (2n = 3x = 21 chromosomes), corresponding to two morphological body patterns, are described. Chromosomes from regenerating blastema were studied after routine Giemsa staining and CBG banding. Our analyses revealed heteromorphisms in chromosomes 2, 3 and 4 of the diploid karyotype and in chromosomes 1, 3 and 7 of the triploid karyotype. Further studies are needed in order to determine if the two morphological patterns of D. carvalhoi represent distinct species.

Comportamento citológico atípico durante a microsporogênese em Brachiaria ruziziensis e B. decumbens. Infoteca-e
RISSO-PASCOTTO, C.; MENDES-BONATO, A. B.; PAGLIARINI, M. S.; VALLE, C. B. do.

Cryptic species of the genus Pimelodella (Siluriformes: Heptapteridae) from the Miranda River, Paraguay River basin, Pantanal of Mato Grosso do Sul, Central Brazil Neotropical Ichthyology
Souza-Shibatta,Lenice; Pezenti,Larissa Forim; Ferreira,Dhiego Gomes; Almeida,Fernanda Simões de; Sofia,Silvia Helena; Shibatta,Oscar Akio.
Specimens of Pimelodellacaptured in the Miranda River, Pantanal of Mato Grosso do Sul State, present morphological features that could indicate at least four species. Therefore, karyotype analysis and molecular biology provided evidence that they were only two species, one showing 2n = 46, and the other, 2n = 52 chromosomes, with only 18% genetic similarity. The morphological analysis evidenced that the dorsal filament is a male characteristic and that the upper lobe of the caudal fin was variable and might or might not be elongated in both species. With respect to morphometric characters, the formation of two groups was evident, but with a small overlap of specimens between them. Among the species with filaments on the dorsal fin observed in the Pantanal,...

Cytogenetic analysis of the Amazon stingless bee Melipona seminigra merrillae reveals different chromosome number for the genus Scientia Agricola
Francini,Izaura Bezerra; Gross,Maria Claudia; Nunes-Silva,Carlos Gustavo; Carvalho-Zilse,Gislene Almeida.
Cytogenetic analysis of the Amazon stingless bee Melipona seminigra merrillae, by conventional Giemsa staining and C-banding, revealed a different chromosome number for Melipona: 2n = 22 for females and diploid drones while the haploid drones present n = 11. There is no evidence of B chromosomes. This result contrasts with previous studies, in which the chromosome number of 19 Melipona species was determined as 2n = 18 for females and n = 9 for haploid males. Based on cytogenetic information available for other Melipona species, we propose that M. s. merrillae has a more derived diploid number. This indicates that chromosome number is not a conservative characteristic within the genus as previously thought. Cytogenetic data for stingless bees are scarce,...

Cytogenetic analysis of three sea catfish species (Teleostei, Siluriformes, Ariidae) with the first report of Ag-NOR in this fish family Genet. Mol. Biol.
Nirchio,Mauro; Martinez,Emanuel Ricardo Monteiro; Foresti,Fausto; Oliveira,Claudio.
Despite their ecological and economical importance, fishes of the family Ariidae are still genetically and cytogenetically poorly studied. Among the 133 known species of ariids, only eight have been karyotyped. Cytogenetic analyses performed on Genidens barbus and Sciades herzbergii revealed that both species have 2n = 56 chromosomes and Cathorops aff. mapale has 2n = 52 chromosomes: Genidens barbus has 10 Metacentrics (M), 14 Submetacentrics (SM), 26 Subtelocentrics (ST), and 6 Acrocentrics (A), Sciades herzbergii has 14M, 20SM, 18ST and 4A, whereas Cathorops aff. mapale has 14M, 20SM, and 18ST. The nucleolus organizer regions (NORs) were found in a single chromosome pair on the short arm of a large-sized ST pair in Genidens barbus and on the short arm of...

Cytogenetic and molecular analyses in troglobitic and epigean species of Pimelodella (Siluriformes: Heptapteridae) from Brazil Neotropical Ichthyology
Dazzani,Bianca; Garcia,Caroline; Peixoto,Marilena; Trajano,Eleonora; Almeida-Toledo,Lurdes Foresti de.
Samples from seven different locations of the genus Pimelodella were genetically examined, two caves (exclusively subterranean, upper Tocantins River and São Francisco River) and five epigean (from upper Paraná River basin). Cytogenetic analyses revealed the same diploid number (2n=46) for all species besides similarities in both number and location of nucleolar organizer regions and C bands. FISH with 5S rDNA probes and CMA3 staining indicated significant differences among the studied species. Application of PCR-RFLP in ATPase 6 and 8 mitochondrial genes allowed building a minimum evolution phenogram identifying the close evolutionary relationship among groups. Both chromosomal and molecular data were useful to infer the relationships among studied...

Cytogenetic characterization of F1, F2 and backcross hybrids of the Neotropical catfish species Pseudoplatystoma corruscans and Pseudoplatystoma reticulatum (Pimelodidae, Siluriformes) Genet. Mol. Biol.
Prado,Fernanda Dotti do; Nunes,Tatiana Leite; Senhorini,José Augusto; Bortolozzi,Jehud; Foresti,Fausto; Porto-Foresti,Fábio.
The cytogenetic characteristics of Pseudoplatystoma corruscans and Pseudoplatystoma reticulatum and their F1, F2 and backcross hybrids were assessed by using chromosome banding techniques. The diploid number of 56 chromosomes was constant in all species and lineages, with a karyotypic formula containing 20 metacentric, 12 submetacentric, 12 subtelocentric and 12 acrocentric chromosomes. Nucleolar organizer regions (NORs) were identified in two subtelocentric chromosomes in the parents and hybrids, with partial nucleolar dominance in F1 and F2 specimens. Heterochromatic blocks were detected in the terminal and centromeric regions of some chromosomes in all individuals. For parental and hybrid lineages, 18S ribosomal clusters corresponding to NORs and 5S...

Cytogenetic damage related to low levels of methyl mercury contamination in the Brazilian Amazon Anais da ABC (AABC)
AMORIM,MARÚCIA I. M.; MERGLER,DONNA; BAHIA,MARCELO O.; DUBEAU,HÉLÈNE; MIRANDA,DANIELA; LEBEL,JEAN; BURBANO,ROMMEL R.; LUCOTTE,MARC.
The mercury rejected in the water system, from mining operations and lixiviation of soils after deforestation, is considered to be the main contributors to the contamination of the ecosystem in the Amazon Basin. The objectives of the present study were to examine cytogenetic functions in peripheral lymphocytes within a population living on the banks of the Tapajós River with respect to methylmercury (MeHg) contamination, using hair mercury as a biological indicator of exposure. Our investigation shows a clear relation between methylmercury contamination and cytogenetic damage in lymphocytes at levels well below 50 micrograms/gram, the level at which initial clinical signs and symptoms of mercury poisoning occur. The first apparent biological effect with...

Velloso,E.D.R.P.; Chauffaille,M.L.; Peliçario,L.M.; Tanizawa,R.S.S.; Toledo,S.R.C.; Gaiolla,R.D.; Lopes,L.F..
Myelodysplastic syndromes (MDS) and juvenile myelomonocytic leukemia (JMML) are rare hematopoietic stem cell diseases affecting children. Cytogenetics plays an important role in the diagnosis of these diseases. We report here the experience of the Cytogenetic Subcommittee of the Brazilian Cooperative Group on Pediatric Myelodysplastic Syndromes (BCG-MDS-PED). We analyzed 168 cytogenetic studies performed in 23 different cytogenetic centers; 84 of these studies were performed in patients with confirmed MDS (primary MDS, secondary MDS, JMML, and acute myeloid leukemia/MDS+Down syndrome). Clonal abnormalities were found in 36.9% of the MDS cases and cytogenetic studies were important for the detection of constitutional diseases and for differential diagnosis...

Leitao, Alexandra; Chaves, Raquel; Santos, Sara; Boudry, Pierre; Guedes Pinto, Henrique; Thiriot Quievreux, Catherine.
Chromosome preparations of the Olympia oyster Ostrea conchaphila Carpenter were studied using conventional Giemsa, silver staining, and C-banding techniques. The karyotype consists of six metacentric (1, 2, 4, 6, 8, and 10) and four submetacentric (3, 5, 7, 9) chromosome pairs. The silver-stained nucleolus organizer regions (Ag-NORs) were terminally located on the short arms of the submetacentric pair 5 (56% of cases) and on the long arms of submetacentric pair 7 (6% of cases). Constitutive heterochromatin was observed as telomeric C-bands on the short arm of the NOR-bearing chromosome pair 5 and as centromeric blocks of several chromosome pairs. Comparative analysis of patterns of karyotype, Ag-NORs, and C-bands of this species and of five other flat...

Cytogenetics of the Brazilian Bolitoglossa paraensis (Unterstein, 1930) salamanders (Caudata, Plethodontidae) Genet. Mol. Biol.
Silva,Jéssica Barata da; Suárez,Pablo; Nagamachi,Cleusa Yoshiko; Carter,Timothy Frederick; Pieczarka,Julio Cesar.
Plethodontid salamanders of genus Bolitoglossa constitute the largest and most diverse group of salamanders, including around 20% of living caudate species. Recent studies have indicated the occurrence of five recognized species in the Brazilian Amazon Rainforest. We present here the first cytogenetic data of a Brazilian salamander, which may prove to be a useful by contribution to the cytotaxonomy of the genus. Specimens were collected near the "type" locality (Utinga, Belém, PA, Brazil). Chromosomal preparations from duodenal epithelial cells and testes were subjected to Giemsa staining, C-banding and DAPI/CMA3 fluorochrome staining. All specimens showed a karyotype with 13 bi-armed chromosome pairs (2n = 26). Nucleolar Organizer Regions, evidenced by...

Cytogenetics of the freshwater cyclopoid mesocyclops longisetus longisetus (crustacea, copepoda) from São Carlos, São Paulo, Brazil Biota Neotropica
Silva,William Marcos da; Tundisi,Takako Matsumura.
A cytogenetic study was carried out on the early embryonic stages of the cyclopoid species Mesocyclops longisetus longisetus (Crustacea, Copepoda) from freshwater environments of the São Carlos area of south-east Brazil. Chromatin was staining using acetic orcein solution. The species showed 2n = 14 chromosomes and presented chromatin diminution in the 4th embryonic cleavage. The eliminated chromatin was concentrated in the equatorial region of the embryonic cell. The results were compared with data for other cyclopoid species of temperate regions.

Cytotaxonomy, heterochromatic polymorphism and natural triploidy of a species of Astyanax (Pisces, Characidae) endemic to the Iguaçu river basin BABT
Kantek,Daniel Luis Zanella; Noleto,Rafael Bueno; Fenocchio,Alberto Sérgio; Cestari,Marta Margarete.
Cytogenetic analysis with Astyanax sp. D revealed a karyotype of 2n=50 with 2M+26SM+6ST+16A, besides a triploid specimen showing 2n=75 chromosomes (3M+39SM+9ST+24A). C-banding strongly stained the terminal regions of several SM-ST-A chromossomes. Two pairs of acrocentric chromosomes presented interstitial heterochromatin, this state being polymorphic and occuring due to possible paracentric inversions. The results obtained with the AluI restriction enzyme and A3 chromomycin were similar to the C-banding. Relationships were proposed between Astyanax sp. D and A. scabripinnis, as well as considerations for a possible origin of the triploid specimen (2n=3x=75). When comparing the present results with cytogenetic features of other endemic Astyanax species in...

Cytotaxonomy of Parodon nasus and Parodon tortuosus (Pisces, Characiformes): A case of synonymy confirmed by cytogenetic analyses Genet. Mol. Biol.
Bellafronte,Elisangela; Margarido,Vladimir Pavan; Moreira-Filho,Orlando.
Morphological and cytogenetical studies were carried out on the freshwater fish Parodon nasus and Parodon tortuosus in order to evaluate a putative synonymy. The diploid chromosome number observed in both species was 2n = 54 (48M/SM and 6ST) with no differences between the sexes. Despite slight differences in the pattern of heterochromatin distribution and the number of cusps in symphysean teeth, the location of nucleolar organizer regions (NORs) and 5S rRNA genes (both species-specific features) were similar in both species. The remarkable similarity observed between these allopatric species supports recent taxonomical reviews indicating that P. tortuosus is a synonym for P. nasus.
